Test Principle and Methodology
HIV-1 was initially identified as the primary pathogen for AIDS and related conditions, while HIV-2 was discovered in 1986 and has also been associated with AIDS cases worldwide. Both viruses share similar morphological characteristics and transmission modes, with approximately 60% genetic homology in conserved genes.
The HIV-1/HIV-2 ELISA is specifically designed to detect antibodies for both virus types. While cross-reactivity occurs between core proteins, envelope proteins remain type-specific. Initial reactive samples must undergo retesting with alternative HIV-1/HIV-2 EIA kits, with confirmatory testing through immunoblotting, immunofluorescence, or radioimmunoprecipitation required for definitive diagnosis.
